A Simple System for In-droplet Incubation and Quantification of Agglutination Assays
نویسندگان
چکیده
This work reports on a simple system for quantitative sensing of a target analyte based on agglutination in microchannels. Functionalized microbeads and analyte with no prior incubation are flowed in droplets (~2μL) through a thin silicone tube filled with mineral oil at a flow rate of 150 μL/min. Hydrodynamic forces alone produce a highly efficient mixing of the beads within the droplet, without the need of complex mixing structures or magnetic actuation. The setup allows rapid observation of agglutination (<2 min), which is quantified using image analysis, and has potential application to high-throughput analysis.

Development and evaluation of realistic microbioassays in freely suspended droplets on a chip.
A novel technique for biomolecular detection in microliter droplets floating on the surface of high density oil is presented. Each droplet was captured and manipulated dielectrophoretically and was used as a site for a microscopic bioassay based on agglutination of antibody-conjugated particles. The results were read out by the pattern of unagglomerated gold nanoparticles collected on the dropl...
متن کاملComparison of Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR) and Agglutination Assays in Diagnosis of Brucellosis in Golestan Province, North of Iran
Introduction: Brucellosis is one of the most common zoonotic infections worldwide. The clinical symptoms of brucellosis are similar to a wide range of diseases; hence, reliable diagnostic and laboratory methods are required to identify the causative agent. Iran is an endemic region of brucellosis, and many patients are misdiagnosed due to the nature of the infection. In this study, we aimed to ...
متن کاملA low cost and high throughput magnetic bead-based immuno-agglutination assay in confined droplets.
Although passive immuno-agglutination assays consist of one step and simple procedures, they are usually not adapted for high throughput analyses and they require expensive and bulky equipment for quantitation steps. Here we demonstrate a low cost, multimodal and high throughput immuno-agglutination assay that relies on a combination of magnetic beads (MBs), droplets microfluidics and magnetic ...
متن کاملQuantification Analysis of Dot Blot Assays for Human Immunodeficiency Virus Type 1 and 2 Antibodies
Objective Dot Blot (DB) assay provides highly specific results, but usually not reliable for quantification of antibody production. The need for a more objective DB assay to provide a better definition of the immune status, against HIV antigens, promoted this study to be done to develop a quantitative DB assay. Material and Methods Dot blot (DB) strips for antibodies directed to human immuno...

Background and purpose: Leptospirosis is a common zoonosis throughout the world and common in the flat area of Guilan, Iran, with seasonal incidence, especially in rice farmers. Clinical diagnosis of leptospirosis is difficult, because its symptoms are similar to several acute infective diseases. Serological assays are important in diagnosis of the disease and microscopic agglutination test (...
